Analysis of iPSC Lines for Alzheimer's Research: KOLF2.1J 

This study characterized widely available induced pluripotent stem cell (iPSC) lines and identified KOLF2.1J as a line that performs well in multiple experimental approaches. Researchers analyzed transcriptional patterns in the pluripotent state, whole genome sequencing, genomic stability after highly efficient CRISPR-mediated gene editing, integrity of the p53 pathway, and differentiation efficiency towards multiple lineages   

Across every measure, KOLF2.1J performed well, making it the reference line selected to anchor NIH's iPSC Neurodegenerative Disease Initiative (iNDI), run through the Center for Alzheimer's and Related Dementias (CARD).  

This research was supported in part by the Intramural Research Program of the NIH, National Institute on Aging (NIA), Department of Health and Human Services, and the National Institute of Neurological Disorders and Stroke (NINDS). 

What the Dataset Contains

The dataset contains:

  • Whole genome and single-cell RNA sequencing files from the analysis 
  • Temporal proteomic profiling of KOLF2.1J-derived neurons across a 28-day differentiation course (n = 6), sampled at seven time points: day 0, 3, 7, 10, 14, 21, and 28 
  • Anchors the NIH CARD-led iPSC Neurodegenerative Disease Initiative (iNDI) 
  • Catalogue entry last updated September 8, 2025; original characterization published December 14, 2021 

What Researchers Can Do With This Dataset

This dataset supports research into the genetic makeup of early-stage Alzheimer's disease, including studies of relatedness and ancestry within the cohort, and analyses that combine genotype data with the LCS's cognitive, clinical, and neuroimaging data. Independent imputation panels support researchers working across different reference populations or replication studies.

The KOLF2.1J iPSC Line Analysis for Alzheimer's Research dataset is discoverable through the AD Discovery Portal and accessible within individually permissioned AD Workbench workspaces at no cost to qualified researchers. AD Workbench provides free compute, virtual machines, and multimodal analysis tools and allows researchers to bring their own code, models, and approved external datasets into their workspace.
